Solution:

pBR 322 has two selectable markers which are amphicillin and tertacyclin resistant genes. Amphicilin resistant gene has two restriction enzyme recognition site, Pvu I and Pst I. If the vector is cut by Pvu I enzyme for formation of rDNA than amphicillin resistant gene becomes inactive due to insertional inactivation.
