DNA fragments are negatively charged and when an electric current is applied they move towards the anode (positively charged electrode). The DNA fragments separate from each other based on their size. The lightest DNA fragment will move faster towards the anode while the heaviest will lag behind. So the one closest to cathode (away from the anode) has to be heaviest and negatively charged.
