All compounds containing peptide linkage (−CO⋅NH−) give respond to Biuret test. For example, urea and proteins. Biuret test for urea: On heating, urea loses NH3 and forms solid biuret. This is dissolved in the least amount of water and made alkaline by adding a few drops of sodium hydroxide. The alkaline solution, when treated with 1−2 drops of dilute CuSO4 solution, gives a violet colour. This test is known as Biuret test.
