The cloning vectors should have a) an Ori which is the origin of replication. b) a Selectable marker c) cloning sites. The commonly used vectors are E coli, plasmids, etc. Artificially prepared vectors are puC and pBr322.If one wants to recover many copies of the target DNA it should be cloned in a vector whose origin support high copy number. But cloning vectors with low copy numbers can also be used. High copy number is not absolutely necessary.
