Solution:

An active site of an enzyme is a crevice or pocket into which the substrate fits. Thus, enzymes catalyse reactions at a high rate through their active site. Once the site is blocked, the enzyme-substrate reaction is stopped, as the substrate cannot complete the reaction without forming its complex with the enzyme at the active site.
