The deletion or addition of a single nucleotide in a coding strand of a gene results in an altered reading frame in the mRNA. The machinery translating the mRNA does not recognize that a base is added or missing, because there is no punctuation in the reading of codons. Thus a major alteration in the sequence of polymerized amino acids results.
